Gelatin-based micro-droplet standards\u00a0(not to be confused with liquid microdroplets\u00a0used\u00a0for single-particle\/cell standardization*)\u00a0were produced by micro-spotting\u00a0of\u00a0exact standard volumes\u00a0onto glass slides,\u00a0resulting in droplet sizes of 200\u00a0\u03bcm\u00a0in diameter.\u00a0A low dispersion laser ablation setup and a\u00a0TOFWERK\u00a0<a href=\"https:\/\/www.tofwerk.com\/zh\/chanpin-2\/icptof\/\">icpTOF<\/a> 2R were used for\u00a0high-throughput\u00a0LA-ICP-TOFMS enabling multi-elemental imaging at a rate of 200 pixels per second.\u00a0Importantly, selective ablation of the gelatin micro-droplets\u00a0needs to be achieved\u00a0by selecting a\u00a0laser\u00a0energy\u00a0density below the ablation threshold of\u00a0the\u00a0glass and above the\u00a0thresholds\u00a0of gelatin and the samples.\u00a0<\/p>\n\n\n\n<p>Analytical&nbsp;figures of merit&nbsp;of this&nbsp;new&nbsp;calibration approach,&nbsp;including&nbsp;limits&nbsp;of detection, precision, and accuracy,&nbsp;were assessed for platinum and&nbsp;other&nbsp;elements with biological key functions, such as&nbsp;phosphorus, copper,&nbsp;and&nbsp;zinc.&nbsp;The applicability of the technique was demonstrated on tumor tissue samples containing&nbsp;metal-based anticancer drugs&nbsp;but can certainly be extended to a range of other bioimaging applications.&nbsp;<\/p>\n\n\n\n<p>*Note by TOFWERK&nbsp;<\/p>","protected":false},"excerpt":{"rendered":"<p>Micro-Droplet-Based Calibration for Quantitative Elemental&nbsp;Bioimaging by LA-ICPMS Schweikert et al.Analytical and Bioanalytical Chemistry,&nbsp;2021DOI:10.1007\/s00216-021-03357-w In this work, researchers from the University of Vienna present a novel\u00a0\u2013\u00a0potentially groundbreaking\u00a0\u2013\u00a0standardization technique for quantitative elemental bioimaging by laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS).
